BACKGROUND: Melanoma is an aggressive malignant tumor, rarely observed in the oral cavity. The aim of this study was to describe the clinicopathologic features of a series of oral melanomas. MATERIAL AND METHODS: A retrospective descriptive study was performed. A total of 15,482 biopsy records from two oral and maxillofacial pathology services in Brazil were analyzed. All cases of oral melanomas were reviewed, and clinical, demographic, histopathological data, treatment, and follow-up status were collected. In addition, immunohistochemistry stains (pan-cytokeratin AE1/AE3, vimentin, α-SMA, CD45, S-100 protein, HMB-45, Melan A, and Ki-67) were performed. RESULTS: The series comprised of 5 males (71.4%) and 2 females (28.6%), with a mean age of 58.0 ± 9.2 years (range: 45-69 years) and a 2.5:1 male-to-female ratio. The gingiva (n = 3, 42.8%) and hard palate (n = 2, 28.6%) were the most common affected sites, presenting clinically as ulcerated swellings with a brown to black color. Cervical lymph node metastasis was detected in three patients during the first examination. Microscopically, 6 cases (85.7%) were melanotic, and one (14.3%) was amelanotic. Most cases (n = 4, 57.1%) presented a predominance of epithelioid cells. S-100 and HMB-45 were positive in all cases (n = 7, 100.0%). In contrast, only 4 cases (57.1%) were positive for Melan-A. The proliferative index with Ki-67 was high, with labeling index ranging from 70.0% to more than 90% of positive cells. Five patients died from complications of the tumors after a mean follow-up period of 7.8 months. CONCLUSIONS: Melanoma is an aggressive malignant tumor that rarely occurs in the oral cavity. It occurs mainly in adult and elderly patients and often is diagnosed in advanced stages. The current findings were similar to previous studies and reflected the characteristics of the services from where lesions were retrieved.

BACKGROUND: Recent studies show that most systemic mastocytosis (SM) patients, including indolent SM (ISM) with (ISMs+) and without skin lesions (ISMs-), carry the KIT D816V mutation in PB leukocytes. We investigated the potential association between the degree of involvement of BM hematopoiesis by the KIT D816V mutation and the distribution of different maturation-associated compartments of bone marrow (BM) and peripheral blood (PB) CD34+ hematopoietic precursors (HPC) in ISM and identified the specific PB cell compartments that carry this mutation. METHODS: The distribution of different maturation-associated subsets of BM and PB CD34+ HPC from 64 newly diagnosed (KIT-mutated) ISM patients and 14 healthy controls was analyzed by flow cytometry. In 18 patients, distinct FACS-purified PB cell compartments were also investigated for the KIT mutation. RESULTS: ISM patients showed higher percentages of both BM and PB MC-committed CD34+ HPC vs controls, particularly among ISM cases with MC-restricted KIT mutation (ISMMC ); this was associated with progressive blockade of maturation of CD34+ HPC to the neutrophil lineage from ISMMC to multilineage KIT-mutated cases (ISMML ). Regarding the frequency of KIT-mutated cases and cell populations in PB, variable patterns were observed, the percentage of KIT-mutated PB CD34+ HPC, eosinophils, neutrophils, monocytes and T cells increasing from ISMs-MC and ISMs+MC to ISMML patients. CONCLUSION: The presence of the KIT D816V mutation in PB of ISM patients is associated with (early) involvement of circulating CD34+ HPC and multiple myeloid cell subpopulations, KIT-mutated PB CD34+ HPC potentially contributing to early dissemination of the disease.

Amblyomma sculptum (Ixodida: Ixodidae) Berlese, 1888, a member of the Amblyomma cajennense complex, is the major vector of Brazilian spotted fever (BSF) in southeastern Brazil. In this study, the genetic diversity of A. sculptum populations in the state of Rio de Janeiro (RJ), Brazil, was investigated because genetic variability in tick populations may be related to vector competence. Samples of A. sculptum from 19 municipalities in 7 regions of RJ were subjected to DNA extraction, amplification and sequencing of D-loop, cytochrome oxidase II and 12S rDNA mitochondrial genes. These sequences were used to map the genetic diversity of this tick. Amblyomma sculptum populations are genetically diverse in RJ, especially in the South Centre and Highland regions. Few unique haplotypes were observed in all populations, and the majority of genetic variation found was among ticks within each population. Phylogenetic reconstruction reinforced the assumption that all the haplotypes identified in RJ belong to A. sculptum. However, some RJ haplotypes are closer to A. sculptum from Argentina than to A. sculptum from elsewhere in Brazil. In RJ, A. sculptum has high genetic diversity, although little genetic differentiation. Observations also indicated a high level of gene flow among the studied populations and no evidence of population structure according to region in RJ.

Cacajao calvus ucayalii (Thomas, 1928) (Primates: Pitheciidae), a subspecies endemic to the Peruvian Amazon, occurs in patchy and sometimes isolated populations in north-eastern Peru and is in a vulnerable situation, mainly due to habitat loss and hunting. This rareness and remote distribution means that, until now, parasitical studies have been limited. Based on optical and scanning electron microscopy of specimens of both sexes, we report two new species of Trypanoxyuris pinworms occurring in the large intestine of the Peruvian red uakari, namely Trypanoxyuris (Trypanoxyuris) cacajao and Trypanoxyuris (Trypanoxyuris) ucayalii. Both species showed a distinct morphology of the lips and cephalic structure. Sexual dimorphism in the lateral alae was observed in both male and the female worms, with ventral ornamentation being shown in the oesophageal teeth. The finding of these new pinworm species highlights the possibility of discovering other species.

RESUMO Boldo-do-chile (Peumus boldus) e camomila (Matricaria recutita) são plantas empregadas na fitoterapia principalmente para o tratamento de desordens hepáticas e intestinais, atuando também como anti-inflamatório e antiespasmódico, respectivamente. Por isso, o presente trabalho objetivou avaliar a qualidade farmacognostica dessas drogas vegetais comercializadas em Fortaleza (CE), além das informações de suas rotulagens. Quarenta e duas amostras de boldo, e quarenta e cinco amostras de camomila, procedentes de farmácias, de lojas de produtos naturais e de bancas de raizeiros, foram analisadas quanto à autenticidade, à pureza e às informações contidas na rotulagem desses produtos conforme legislação vigente. Na verificação de impurezas, 35,7% das amostras de boldo e 57,7% das amostras de camomila excederam o teor máximo de matéria estranha; no teor de cinzas totais, 33,3% das amostras de boldo foram reprovadas; rotulagens das amostras de boldo (100%) e de camomila (96,6%) apresentaram erros ou ausência de informações. Os resultados das análises confirmam, portanto, a necessidade urgente de melhor fiscalização e intervenção na produção e venda dessas e de outras drogas vegetais para adequação às normas vigentes.


ABSTRACT Boldo (Peumus boldus) and chamomile (Matricaria recutita) plants are mainly used in herbal medicine to treat hepatic and gastrointestinal diseases, having also anti-inflammatory and antispasmodic properties. This study aimed to evaluate the pharmacognostic quality of these herbal drugs sold in Fortaleza (CE), besides analyzing the information on their labeling. Forty-two samples of boldo, and forty-five samples of chamomile, brought from pharmacies, health food stores and newsstands were analyzed regarding their authenticity, purity, and the information contained in the labeling of the industrial products, to assure that it was in accordance with the current legislation. When verifying the impurities, 35.7% of the boldo samples and 57.7% of the chamomile ones exceeded the maximum level of strange organic matter allowed. Concerning the total ash content, 33.3% of boldo samples were rejected. The labels on the packages of boldo (100%) and chamomile (96,6%) contain errors or lack of information. The test results confirm the need for better surveillance and intervention in the production and sale of these and other drugs plants in order to meet the current standards.

Durante 24 meses foram capturados, inspecionados e liberados no mesmo espaço do Parque Estadual da Pedra Branca, Rio de Janeiro, 96 marsupiais e 64 roedores. Neles foram recolhidos manualmente 105 carrapatos, de 10 espécies em duas famílias. A espécie de carrapato dominante entre os roedores foi Amblyomma longirostre e entre os marsupiais foi Ixodes loricatus. Houve correlação direta significativa entre a temperatura e a intensidade de parasitismo por carrapatos...


During 24 months 96 marsupials and 64 rodents were captured, inspected and set free in the same space of the Pedra Branca State Park, Rio de Janeiro. From them, 105 ticks from 10 species in two families were manually collected. The dominant tick species on the rodents was Amblyomma longirostre and on the marsupials was Ixodes loricatus. There was a significant correlation of the temperature in relation to intensity of parasitism for ticks, demonstrating that the higher the temperature, greater is the number of ticks...

RESUMO

Eight wild fowls kept in captivity at a Brazilian Zoo were examined from july 1994 to October 2000. One hundred twenty-three Ixodidae specimens were collected and sent to the Ixodides Laboratory at the Instituto Oswaldo Cruz, Rio de Janeiro, Brazil. They were examined by stereomicroscopy and were identified as Amblyomma longirostre (53), A. pacae (50), A. cajennense (14), and A. parvum (6).

The properties of granite powders--a granite manufacturing waste product-were analyzed to assess their potential use as amendments and fertilizers on acid soils. Two types of powders were characterized: one produced during cutting of granite with a diamond-edged disc saw, comprising only rock powder, the other produced during cutting with a multi-blade bandsaw, containing calcium hydroxide and metal filings added during the cutting procedure. The acid neutralizing capacity of the granite powders was assessed in short- (2-3 h) and medium-term (1-30 d) experiments. The powders showed a buffering capacity at around pH 8, which corresponded to the rapid dissolution of basic cations, and another buffering effect at pH<4.5, attributable to the dissolution of Fe and Al. The acid neutralizing capacity (ANC) determined in the short-term experiments, to a final pH of 4.5, varied between 5 and 61 cmol H+kg(-1) powder. The ANC to pH 4.5 obtained in the medium-term experiments was much higher than that obtained in the short-term experiments, reaching a maximum ANC value of 200 cmol H+kg(-1) powder. There was no great difference in the neutralizing capacity determined at between 1 and 30 d. The most abundant elements in acid solutions obtained at the end of medium-term experiments were Mg and Ca for disc saw powders, whereas Ca and Fe (at pH<5) were the most soluble elements in the bandsaw powders. The rapid release of these cations suggests the possible effective use of the granite powders as a source of nutrients on being added to acid soils.

RESUMO

Larvae of Amblyomma cajennense (Fabricius, 1787), were obtained under laboratory conditions. The larvae were killed in hot water, preserved in 70° ethanol and prepared in definitive whole mounts. Two hundred larvae were examined under brightjield microscopy with an immersion objective in order to analyze the variation of the number of internal setae of the right and left side of the Haller's organ chamber with the purpose of utilize them as chaetotaxy parameter for specific diagnosis.

Eggs were obtained from female Argas (Persicargas) miniatus parasitizing chickens after natural ovoposition. The larvae from eggs were sacrificed, prepared for and mounted onto slides. Morphological and chaetotaxic studies enabled the following observation: idiosome and gnathosome shape; presence or absence of scutum, eyes and anal setae; number of palpi segments; number of files of teeth on the hypostoma; number of denticles on the apical crow; number of cheliceral digits; number of teeth in each hypostoma file; disposition and shape of opistosoma and podosoma setae in ventral and dorsal view. Based on these morphological features analyzed, it is concluded that the larvae of A. (P.) miniatus present variations in morphology and chaetotaxy that are useful to separate the larvae of the subgenus Persicargas.

OBJECTIVE: To determine the duration of the outpatient diagnostic process for lung cancer in comparison to that of other solid organ tumors/all tumors at the National Oncology Institute-Society to Fight Cancer (ION-SOLCA) of Ecuador. PATIENTS AND METHODS: All patients with non-small cell lung cancer (NSCLC) seen between January 1 and December 31, 1995 at the ION-SOLCA, a specialized tertiary care hospital in Guayaquil, Ecuador, were studied. The duration of the patients' diagnostic process was compared to that of other patients with solid organ tumors (1 control per NSCLC patient). DESIGN: Retrospective study of health care services to measure the duration of each stage of the diagnostic process for cancer patients at the ION-SOLCA. MEASURES: The main variable was the duration of the diagnostic process. The duration of each phase of the process was also recorded. RESULTS: Results are given as means ( standard deviations, with standard errors between parentheses). The overall duration of the diagnostic process for all solid organ tumors (lung and others) at the ION-SOLCA was 54.5 days 62.3 (7.6). No differences were detected between the duration of diagnosis for lung and other tumors. The durations of the different phases of diagnosis were as follows: from the first pre-admission contact with the hospital until a visit with a specialist, 12.5 days 11.4 (1.4); from the visit with a specialist until a diagnostic procedure, 33.3 days 57 (7); and from the diagnostic procedure until the pathological diagnosis, 8.7 days 6.9 (0.8). CONCLUSIONS: Outpatient evaluation is an inefficient, slow and potentially dangerous process in cases in which the probability of a cancer diagnosis is high. A more interventionist process involving hospital admission may accelerate diagnosis in such cases.

OBJETIVO: Determinar la duración del proceso diagnóstico del cáncer (pulmón frente a tumores de órganos sólidos/todos) en el ION-SOLCA utilizando la modalidad de evaluación ambulatoria. POBLACIÓN Y MÉTODOS: La totalidad de los pacientes con cáncer de pulmón de células no pequeñas (CPCNP) evaluados durante el período comprendido entre el 1 de enero y el 31 de diciembre de 1995 en el ION-SOLCA de Guayaquil, Ecuador (hospital de referencia terciario), comparando con la duración de la evaluación diagnóstica de otras neoplasias de órganos sólidos en la institución. DISEÑO: Estudio retrospectivo de servicios de salud que cuantifica la duración de cada etapa del proceso diagnóstico de los pacientes con cáncer en el ION-SOLCA. Se consideró como variable índice del estudio la duración de la evaluación diagnóstica de los casos estudiados en el ION-SOLCA. Simultáneamente se obtuvo la duración de cada componente de la variable índice. RESULTADOS: La duración media de la evaluación diagnóstica global en el ION-SOLCA para los tumores de órganos sólidos (pulmón y otros) es de 54,5 ñ 62,3 días (error estándar [EE], 7,6). No se encontró diferencia entre los dos grupos. Los componentes de esta estimación se fraccionan del siguiente modo: primer contacto con el instituto (preadmisión)-visita con el especialista, 12,5 ñ 11,4 días (EE, 1,4); visita con el especialista-procedimiento diagnóstico, 33,3 ñ 57 días (EE, 7), y procedimiento diagnóstico-resultado de anatomía patológica, 8,7 ñ 6,9 días (EE, 0,8).CONCLUSIONES: La valoración ambulatoria de los pacientes con alta probabilidad de cáncer es un proceso ineficiente, lento y posiblemente peligroso. Una modalidad de ingreso intervencionista podría acelerar el proceso diagnóstico en estos pacientes (AU)

From August 1999 to January 2000, samples of house dust were collected from 160 domiciles in the city of Juiz de Fora, State of Minas Gerais, Brazil. In 36 of these domiciles kitchen samples were obtained. Prevalence rate was 77.5%, varying according to the geographical sector. There were found 2,278 specimens of mites, with 1,530 (67.2%) in the adult stage and 748 (32.8%) in immature forms. The main species found were Dermatophagoides pteronyssinus, D. farinae, Euroglyphus maynei, Blomia tropicalis and Tyrophagus putrescentiae. In a minor incidence we found Lepidoglyphus destructor, Suidasia pontificiae, Chortoglyphus arcuatus, Cheyletus malaccensis, C. fortis, Ker bakeri, Cheletonella vespertilionis, C. caucasica and others. C. vespertilionis and C. caucasica were identified for the first time in the domiciliary ecosystem and in Brazil. The abundance rate and the infestation intensity were analyzed. There was a varied correlation between climatic conditions and positive domiciles and number of mites. The difference between the number of positive domiciles in the urban area and in the expanding urban area was significant and so was the difference between samples from the domiciles compared to those from the kitchens.

Eleven species of fleas were collected from 601 small rodents, from November 1995 to October 1997, in areas of natural focus of bubonic plague, including the municipalities of Nova Friburgo, Sumidouro and Teresópolis, State of Rio de Janeiro, Brazil. Among 924 fleas collected, Polygenis (Polygenis) rimatus (Rhopalopsyllidae) was the predominant species regarding the frequency, representing 41.3% (N:382), followed by P. (Neopolygenis) pradoi, representing 20% (N:185) and Craneopsylla minerva minerva (Stephanocircidae), representing 18.9% (N:175). The host Akodon cursor harbored 47.9% of these fleas. Other six host species were infested by 52.1% of the remaining fleas. Fleas were found on hosts and in places within the focus not previously reported by the literature.

Variation between aeropile numbers of the right and left peritrematic plate in male and female Anocentor nitens (Neumann, 1897) is reported from a site in Brazil. From January to December 1998, 146 males and 247 females of A. nitens were recovered from Equus caballus L. in Silva Jardim District, State of Rio de Janeiro. Asymmetry of numbers of aeropiles between right and left plates occurred in 83.6% of the males and 82.2% of the females. Differences in the number of aeropiles between the sexes were not significant. Quantitative variation of aeropiles was correlated to the period of recovery, with significant asymmetry detected in August-September and November-December, mainly in males. Results suggest an adaptation, especially in the male ticks, that expresses itself as greater variation in the number of aeropiles in some periods of the year.

RESUMO

Mites collected from the auditory canal of Cebus apella (capuchin monkey), family Cebidae, were identified as Fonsecalges johnjadini (Psoroptidae, Cebalginae). It is the first record of this parasite from this monkey. This paper emphasizes the importance of clinical and anatomopathological examinations for parasitic diagnosis in wild animals.

RESUMO

A large body of diverse comparative data now exists for a major phylogenetic synthesis of the higher-level relationships among eutherian (placental) mammals. We present such a phylogenetic synthesis using the composite trees or supertrees from the combined and separate analyses of their published molecular and morphological source phylogenies. Our combined and separate supertrees largely support the same suprafamilial taxa and orders, but different interordinal clades. These similarities and differences reinforce the continuing contributions of morphological studies, while highlighting the growing influence of molecular information on the field. As current summaries of past research, our supertrees emphasize opportunities for future work, while providing a step toward the eventual integration of the data and characters themselves.

CpG islands are discrete regions of DNA with significantly greater frequencies of CpG doublets than bulk genomic DNA. They are most frequently associated with the 5'-ends of housekeeping genes and are involved in the regulation of their expression. In this study, the structure and evolution of CpG islands within genes of the myc family were evaluated with the protein-coding sequences of animals and their transducing viruses. These evaluations relied on a gene tree for the entire myc family to test the origins of CpG islands within their two protein-coding exons. Overall, CG-very rich and CG-rich islands are associated with exon 2 of the different myc genes of warm-blooded vertebrates and with exon 3 of the N-myc and s-myc sequences of mammals, but not birds. These overall distributions of well-developed islands can be related to the major transitions of the CG-rich genomes of warm-blooded vertebrates from the CG-poor ones of other animals. In turn, the greater variability of well-developed islands within exon 3 of the N-myc gene and among the different retrogenes of the myc family can be attributed to their reduced functional constraints, as evidenced by their limited and very restricted patterns of expression, respectively.
